چکیده

Although the regulation of epithelial morphogenesis is essential for the formation of tissues and organs in multicellular organisms, little is known about how signalling pathways control cell shape changes in space and time. In the Drosophila ovarian epithelium, the transition from a cuboidal to a squamous shape is accompanied by a wave of cell flattening and by the ordered remodelling of Ecadherin-based adherens junctions. We show that activation of the TGFb pathway is crucial to determine the timing, the degree and the dynamic of cell flattening. Within these cells, TGFb signalling controls cell-autonomously the formation of Actin filament and the localisation of activated Myosin II, indicating that internal forces are generated and used to remodel AJ and to promote cytoskeleton rearrangement. Our results also reveal that TGFb signalling controls Notch activity and that its functions are partly executed through Notch. Thus, we demonstrate that the cells that undergo the cuboidal-to-squamous transition produce active cell-shaping mechanisms, rather than passively flattening in response to a global force generated by the growth of the underlying cells. Thus, our work on TGFb signalling provides new insights into the mechanisms through which signal transduction cascades orchestrate cell shape changes to generate proper organ structure.

Quantitative analysis of epithelial morphogenesis in Drosophila oogenesis: New insights based on morphometric analysis and mechanical modeling.

The process of epithelial morphogenesis is ubiquitous in animal development, but much remains to be learned about the mechanisms that shape epithelial tissues. The follicle cell (FC) epithelium encapsulating the growing germline of Drosophila is an excellent system to study fundamental elements of epithelial development. During stages 8 to 10 of oogenesis, the FC epithelium transitions between ...

Top–DER- and Dpp-dependent requirements for the Drosophila fos/kayak gene in follicular epithelium morphogenesis

The Drosophila fos (Dfos)/kayak gene has been previously identified as a key regulator of epithelial cell morphogenesis during dorsal closure of the embryo and fusion of the adult thorax. We show here that it is also required for two morphogenetic movements of the follicular epithelium during oogenesis. Firstly, it is necessary for the proper posteriorward migration of main body follicle cells ...

Expression of fringe is down regulated by Gurken/Epidermal Growth Factor Receptor signalling and is required for the morphogenesis of ovarian follicle cells.

Signalling by the Gurken/Epidermal Growth Factor Receptor (Grk/EGFR) pathway is involved in epithelial cell fate decision, morphogenesis and axis establishment in Drosophila oogenesis. In the search for genes downstream of the Grk/EGFR signal transduction pathway (STP), we isolated a number of genes that are components of other STPs. One of them is a known gene, called fringe (fng). Drosophila ...

ERRATUM Capicua regulates follicle cell fate in the Drosophila ovary through repression of mirror

We apologize to the authors and readers for this mistake. INTRODUCTION Epithelial patterning during development involves the generation, transmission and interpretation of positional cues. In the egg chambers of the Drosophila ovary, the follicular epithelium is patterned through a series of localized inductive interactions with At the end of oogenesis, the follicle cells secrete the eggshell, ...
